conclusion: เลิกแต่งกลอนดีกว่า ปวดหัวค่ะ เดี๋ยวมาแต่งใหม่
ขอโทษที่ไม่ค่อยคล้องจองกันเท่าไหร่ แล้วภาษาไทยอาจจะแปลก ๆ

 January 28 Scientists for Better PCRฟังเพลง คลิ๊กที่นี่ and enjoy! มีคนรู้จักส่งเพลงนี้มาทาง e-mail ค่ะ ฟังดูแล้วก็ชอบนะ กำลังใช้เทคนิคนี้ทำวิจัยอยู่พอดีเลยค่ะ ไม่ได้มีเจตนาโฆษณาเครื่องมือของบริษัท Bio-Rad แต่อย่างใดค่ะ ขออธิบายเพิ่มเติมเกี่ยวกับ Dr. Kary Mullis ที่คิด idea เจ๋ง ๆ เช่น เทคนิค PCR ให้มนุษยชาติค่ะ สมควรแล้วที่ได้รับรางวัล Nobel ในปี 1993 เรื่องมันมีอยู่ว่าเมื่อ 25 ปีที่แล้ว ขณะที่ Dr. Kary Mullis ขับรถอยู่ก็เกิดความคิดที่จะใช้ primers 1 คู่ มาครอบเอาเฉพาะส่วนที่เป็น gene ที่ต้องการ แล้วใช้ DNA polymerase ในการต่อสาย DNA แต่ปัญหาก็คือ DNA polymerase ไม่ทนความร้อน ในขั้นที่ต้องใช้ความร้อนในการแยกสาย DNA (ประมาณ 95 องศาเซลเซียส) โดยต้องเติม DNA polymerase ใหม่ทุกครั้งที่มีการแยกสาย DNA ค่ะ (ก็ถ้าเกิดต้องทำ 30 รอบ ต้องเสียเวลาเปิดฝาเติมลงไป 30 ครั้งนะ มันเสียเวลาอ่ะ) อีก 3 ปีต่อมา เขาก็ใช้ DNA polymerase ที่ทนความร้อน ได้มาจากแบคทีเรียที่อยู่ในนำพุร้อน (Thermophilus Aquaticus, Taq) แทนยังไงล่ะคะ ได้ข่าวมาว่ามีหลายแล็ปเค้าเลี้ยงแบคทีเรียชนิดนี้ไว้ใช้เองด้วย ใครมีก็บอกหน่อยละกันอยากลองเลี้ยงไว้ใช้เองบ้างค่ะ ลงเนื้อเพลงไว้ให้ด้วย อยู่ด้านล่างค่ะ

พ่อกับแม่ก็สบายดี บ้านก็รกตามเคย ก็เลยทำความสะอาดบ้านซะหน่อย
ต้อนรับพี่สาวกลับบ้านด้วย พี่สาวมีอาการ jet lag นิดหน่อย
ได้กลับไปไหว้พระ ทำบุญ รดน้ำมนต์ด้วยขอให้พระคุ้มครอง
แล้วก็ทำวิทยานิพนธ์ด้วยความราบรื่น
พระท่านศักดิ์สิทธิ์จริง ๆ นะ กลับมาก็ทำ PCR ออกเลย
ก่อนกลับบ้าน ตอนแรกกลุ้มใจมาก ทำยังไง PCR ก็ไม่ขึ้น นึกว่าไพรเมอร์มันถูก nuclease ย่อยหมด
แต่พอลองมา test กับ positive ของเรา amp. ขึ้นอยู่นะ แผนที่สองคือ template
มีปัญหา ลองเอามา run gel ดูก็ยังมีอยู่ ไม่ได้ degrade แฮะ ก็เลยคิดว่าสงสัยเก็บ DNA
ไว้นาน มี unknow impurity ขึ้นมา เลยสกัด DNA ใหม่ซะเลย แล้วก็จัดการ purified เสร็จ
ทำ PCR ต่อเลย ปรากฏว่ามี product ขึ้นแล้ว ดีใจมากเลยจะได้ทำขั้นต่อไป
